小鼠腹腔巨噬細(xì)胞和雜交瘤細(xì)胞樣品*次在較好的生長(zhǎng)條件下進(jìn)行培養(yǎng)。細(xì)胞生長(zhǎng)在改良的動(dòng)態(tài)細(xì)胞培養(yǎng)系統(tǒng)內(nèi),該系統(tǒng)包含兩個(gè)細(xì)胞反應(yīng)器,每一個(gè)反應(yīng)器中有培養(yǎng)液儲(chǔ)存系統(tǒng),換液系統(tǒng),兩個(gè)細(xì)胞培養(yǎng)室及氣室。氣室中初始?xì)怏w為含8%CO2、25%O2的混合氣體;氣室與培養(yǎng)室之間有一層可以進(jìn)行氣體交換的薄膜相隔。每一反應(yīng)器中有一個(gè)培養(yǎng)室,由滲透泵驅(qū)動(dòng)換液,培養(yǎng)液流速大約為60微升/天,稱(chēng)為流動(dòng)培養(yǎng)室。另一個(gè)非換液室稱(chēng)為靜止培養(yǎng)室。溫控裝置維持培養(yǎng)室內(nèi)溫度約為36±1℃,培養(yǎng)液儲(chǔ)存室溫度約為15±2°C。經(jīng)過(guò)15天的空問(wèn)飛行試驗(yàn)表明我國(guó)自行研制的這一裝置基本適合巨噬細(xì)胞的培養(yǎng),貼附在微載體上的巨噬細(xì)胞在改良的動(dòng)態(tài)培養(yǎng)系統(tǒng)內(nèi)存活良好。裝置還需要進(jìn)一步改進(jìn)和完善。目前,一個(gè)更高水平和更大容量的細(xì)胞反應(yīng)器正在研制中。原核細(xì)胞(prokaryotic cell)沒(méi)有核膜,遺傳物質(zhì)集中在一個(gè)沒(méi)有明確界限的低電子密度區(qū)。DNA為裸露的環(huán)狀分子,通常沒(méi)有結(jié)合蛋白,環(huán)的直徑約為2.5納米,周長(zhǎng)約幾十納米。沒(méi)有恒定的內(nèi)膜系統(tǒng),核糖體為70S型,原核細(xì)胞構(gòu)成的生物稱(chēng)為原核生物(prokaryote),均為單細(xì)胞生物,通常稱(chēng)為細(xì)菌(bacterium)。
